| Bad Abstract
| This paper aims to analyze a recombinant plasmid and its resistance genes. For this purpose, we will conduct four experiments and determine antibiotic resistance genes through the restriction reactions.
| Good Abstract
| The purpose of the experimental stream is to construct and analyze a recombinant plasmid containing ampicillin and kanamycin antibiotic resistance genes. The donor organism utilized in these experiment is the E.coli bacteria. The reactants in the experiments are the pKAN and pAMP plasmids. The pAMP plasmid carries an antibiotic gene for penicillin while the pKAN plasmid carries an antibiotic gene for kanamycin. In order to achieve the main purpose of the experimental stream, four experiments are conducted which include Lab 1, Lab 2, Lab 3, and Lab 4. Lab 1 focuses on the recombination of the antibiotic resistance genes through digesting the two plasmids in separate restriction reaction and the ligation of the pKAN and pAMP restriction digests. The ligation products are used to transform E.coli cells in Lab 2. The mixed E.coli population is identified in Lab 3 through replica plating while Lab 4 is concerned with the purification and identification of the recombinant DNA.
